[目的]介绍一种适合于富含多糖、多酚植物材料的RNA提取方法。[方法]采用改进的异硫氢酸胍法提取植物材料的RNA,用紫外分光光度计测定其浓度和纯度,并通过RT-PCR进一步检测其质量。[结果]提取总RNA中的28、18、5 S条带清晰,比例适中,OD260 nm/OD280 nm为1.8~2.0。以提取的总RNA反转录合成的cDNA为模板,能扩增出预期长度的片段,无非特异性产物。[结论]该方法简便易行,成本较低,适合于富含多糖、多酚植物材料的RNA提取。
[Objective] The research aimed to introduce a kind of RNA extraction method suitable for polysaccharide and polyphenol plant material. [Method] The RNA of plant material was extracted by the improved guanidium isothiocyanate method, its concentration and purity were determined by ultraviolet spectrophotometer, and its quality was further detected by RT-PCR. [Result] The bands of 28, 18, 5 S in the total RNA were clear and the proportion was moderate. The OD260 nm / OD280 nm was 1.8 ~ 2.0. Reverse transcription of total RNA extracted from the cDNA synthesized as a template, can amplify the expected length of the fragment, non-specific products. [Conclusion] The method was simple and easy to operate with low cost and suitable for RNA extraction from plant material rich in polysaccharides and polyphenols.
