Objective: To investigate the effect of CD40 / CD40 ligand-binding costimulatory signal on the proliferation and differentiation of stem cells and B cells and the anti-leukemia effect of CD40 ligand. Source: PubMed and Springer databases were searched from January 1995 to December 2005 with articles related to CD40 / CD40 ligand, leukemia stem cells and leukemia. The search term was “CD40, CD40L, Leukemia, Leukemicstemcell” and the limited literature language was English. Data Selection: The data were first reviewed, including the experimental and control groups, including experimental literature, excluding significant randomized trials and clinical studies and reproducibility studies. DATA EXTRACTION: A total of 30 experimental articles about the effect of CD40 / CD40 ligand-binding co-stimulatory signals on the proliferation and differentiation of stem cells and B cells and the anti-leukemia effect of CD40 ligand were collected. Twenty-three were eligible for inclusion and 7 were excluded All for the same study. DATA SYNTHESIS: Totally 23 experiments including the effect of CD40 / CD40 ligand on the proliferation and differentiation of leukemic stem cells and B cells, the prognosis of leukemia patients and the therapeutic effect of CD40 ligand on leukemia patients, and the effect of CD40 / CD40 ligand on leukocyte Stem cells, B cells and leukemia in patients with the impact of the role of analysis. Conclusion: The co-stimulatory signals generated by the combination of CD40 / CD40L can promote the proliferation and differentiation of leukemic stem cells and B cells, which is closely related to the occurrence, development and prognosis of leukemia. CD40 / CD40L plays a key role in immune response and is widely used in immunotherapy of leukemia.
