目的探讨在小鼠精原干细胞体外分离过程中,不同的温度对精原干细胞消化作用的影响。方法在小鼠精原干细胞体外分离过程中,建立不同的消化温度环境,观察消化酶对小鼠精原干细胞的离散消化效果及精原干细胞的活力和损伤情况。结果精原干细胞体外消化过程中,37℃环境下消化酶对精原干细胞的消化作用最快,但对精原干细胞的损伤较重;32~34℃环境下消化酶对精原干细胞的消化作用较弱,但对精原干细胞的损伤作用较小,不同温度之间消化快慢与细胞损伤的差异有统计学意义(P<0.01)。结论在小鼠精原干细胞体外分离过程中,不同的温度对精原干细胞的活力和所获得的具有增殖能力的精原干细胞数量均有重要的影响,32~34℃环境温度为精原干细胞体外分离较适合的温度。
Objective To investigate the effects of different temperatures on the digestion of spermatogonial stem cells during the isolation of mouse spermatogonial stem cells in vitro. Methods In the process of isolation of mouse spermatogonial stem cells in vitro, different digestion temperature environments were established to observe the digestive effects of digestive enzymes on spermatogonial stem cells and the viability and injury of spermatogonial stem cells. Results Digestive enzymes of spermatogonial stem cells were the most digestible in spermatogonial stem cells under the condition of 37 ℃ digestion, but had a greater damage to spermatogonial stem cells. Digestive effects of digestive enzymes on spermatogonial stem cells under 32 ~ 34 ℃ Weak, but the damage of spermatogonial stem cells is less, the difference between the digestion speed and the cell damage is statistically significant (P <0.01). Conclusion In the process of in vitro isolation of spermatogonial stem cells from mice, different temperatures have an important effect on the viability of spermatogonial stem cells and the number of spermatogonial stem cells obtained. Proliferation of spermatogonial stem cells at 32 ~ 34 ℃ Separation of more suitable temperature.
