目的研究十二指肠空肠空置手术(DJB)对2型糖尿病(T2DM)大鼠胰腺胰高血糖素样肽-1受体(GLP-1R)表达的影响,探讨DJB手术后胰腺GLP-1R表达变化与血糖的关系。方法雄性Wistar大鼠随机分为4组,每组8只:Wistar正常大鼠组(W-C);T2DM模型对照组(T2DM-C);T2DM二甲双胍组(T2DM-M);T2DM+DJB手术组(T2DM-DJB)。分别测定各组大鼠术前及术后第2、4、8周空腹血糖、空腹血清GLP-1和空腹血清胰岛素值。术后第8周处死大鼠,RT-PCR检测胰腺GLP-1RmRNA的表达变化。结果 T2DM-DJB、T2DM-M组大鼠术后空腹血糖和空腹血清胰岛素显著降低(P<0.01),空腹血清GLP-1显著升高(P<0.01),与相应时间点T2DM-C组相比均有统计学意义(P<0.01)。T2DM-DJB组大鼠术后空腹血糖、空腹血清GLP-1、空腹血清胰岛素与T2DM-M组大鼠相比无明显差异(P>0.05)。术后第8周,RT-PCR结果显示GLP-1RmRNA在T2DM-DJB、T2DM-M组大鼠胰腺表达水平分别为(0.55±0.03)、(0.53±0.01),明显高于T2DM-C组大鼠GLP-1R mRNA的表达水平(0.41±0.02)(P<0.01)。结论 DJB手术能显著降低T2DM大鼠血糖,可能与术后T2DM大鼠胰腺GLP-1R表达水平升高有关。
Objective To investigate the effect of empty duodenum jejunum (DJB) on the expression of GLP-1R in the pancreas of type 2 diabetes mellitus (T2DM) rats and to investigate the expression of GLP-1R Relationship between changes and blood sugar. Methods Male Wistar rats were randomly divided into 4 groups with 8 rats in each group: Wistar normal rats (WC); T2DM model control (T2DM-C); T2DM metformin (T2DM-M); T2DM + T2DM-DJB). The levels of fasting blood glucose, fasting serum GLP-1 and fasting serum insulin were measured before operation and 2, 4, and 8 weeks after operation respectively. The rats were sacrificed at the 8th week after operation, and the expression of GLP-1R mRNA in the pancreas was detected by RT-PCR. Results Fasting plasma glucose (FBG) and fasting serum insulin were significantly decreased in T2DM-DJB and T2DM-M groups (P <0.01) and GLP-1 in fasting serum was significantly increased (P <0.01) (P <0.01). There was no significant difference in fasting blood glucose, fasting serum GLP-1 and fasting serum insulin between T2DM-DJB group and T2DM-M group (P> 0.05). At the 8th week after operation, the expression of GLP-1R mRNA in the T2DM-DJB and T2DM-M groups was (0.55 ± 0.03) and (0.53 ± 0.01), respectively, which was significantly higher than that in the T2DM-C group The GLP-1R mRNA expression level in rats was (0.41 ± 0.02) (P <0.01). Conclusion DJB can significantly reduce blood glucose in T2DM rats, which may be related to the increased expression of GLP-1R in the pancreas of T2DM rats.
