目的:探讨芍药苷对5-氟尿嘧啶(5-FU)耐药的CD44阳性胃癌细胞株SGC-7901(CD44+SGC-7901/5-FU)细胞增殖与凋亡的影响。方法:采用反复短期暴露并逐步递增药物浓度法建立5-氟尿嘧啶耐药胃癌细胞株SGC7901/5-FU,鼠抗人CD44抗体,流式细胞术检测分选CD44+SGC-7901/5-FU细胞。将不同浓度的芍药苷作用于体外扩增培养的CD44+SGC-7901/5-FU细胞后,分别用倒置显微镜观察细胞形态学改变、MTT法检测细胞的增殖情况、流式细胞仪检测细胞凋亡率。结果:各浓度芍药苷作用于CD44+SGC-7901/5-FU细胞后,细胞形态与对照组细胞比较发生了明显变化;与对照组比较细胞生长明显受到抑制、细胞凋亡率明显增加,且呈明显的时间与浓度依赖性(均P<0.05)。结论:芍药苷能在体外抑制5-FU耐药的人胃癌细胞的生长。
Objective: To investigate the effects of paeoniflorin on proliferation and apoptosis of 5-fluorouracil-resistant CD44-positive gastric cancer cell line SGC-7901 (CD44 + SGC-7901/5-FU). Methods: The gastric cancer cell lines SGC7901 / 5-FU and mouse anti-human CD44 were established by repeatedly short-term exposure and gradually increasing drug concentration. CD44 + SGC-7901/5-FU cells were sorted by flow cytometry . The different concentration of paeoniflorin was used to expand the cultured CD44 + SGC-7901/5-FU cells in vitro. The morphological changes of the cells were observed by inverted microscope. The proliferation of cells was detected by MTT assay. The apoptosis of cells was detected by flow cytometry Death rate. Results: Paeoniflorin at different concentrations significantly changed the morphology of cells compared with the control cells after treated with CD44 + SGC-7901/5-FU cells. Compared with the control group, the cell growth was significantly inhibited and the cell apoptosis rate was significantly increased Significant time and concentration-dependent (P <0.05). CONCLUSION: Paeoniflorin can inhibit the growth of 5-FU resistant human gastric cancer cells in vitro.
