目的:探索家兔类风湿关节炎(RA)寒证模型的建立方法。方法:90只清洁级青紫蓝家兔随机分为空白组、卵蛋白组、卵蛋白+冷冻组,每组30只。卵蛋白组采用卵蛋白溶液与等量弗氏完全佐剂混成乳化剂,在家兔肩背部6个点各注射0.2m L,14d后再注射;卵蛋白+冷冻组注射相同乳化剂,在末次注射24h后冷冻1.5h,复温5min。冷冻后1d随机分为3亚组,观察各亚组冷冻后1、7、14d一般表现、膝周径、关节表面温度及关节滑膜病理积分变化。结果:空白组未见异常;与前者比较,卵蛋白+冷冻组、卵蛋白组膝关节肿胀、滑膜病理积分均增高(P<0.05);与同组冷冻后1d比较,冷冻后7d肿胀达到高峰、滑膜病理积分较高(P<0.05);与同组冷冻后7d比较,冷冻后14d膝关节肿胀减轻、滑膜病理积分减小(P<0.05),但仍较冷冻后1d积分高(P<0.05)。结论:采用卵蛋白诱导联合低温冷冻法建立的RA寒证模型可靠,模型至少可持续14d。
Objective: To explore a method to establish the cold syndrome model of rabbit rheumatoid arthritis (RA). Methods: Ninety clean blue-purple blue rabbits were randomly divided into blank group, ovalbumin group, ovalbumin + frozen group, 30 in each group. The egg albumin solution was mixed with the same amount of Freund’s complete adjuvant to prepare an emulsifier. Each egg was injected into the shoulder and back of the rabbits at a dose of 0.2mL for 14 days and then injected for 14 days. The same emulsifier was injected into the ovalbumin + After 24h injection frozen 1.5h, rewarming 5min. One day after freezing, the rats were randomly divided into three subgroups. The general performance, the knee circumference, the joint surface temperature and the synovial pathological changes of the subgroups were observed at 1, 7, 14 days after freezing. Results: Compared with the former group, the swelling of the knee joint and the synovial pathological score of the ovalbumin + frozen group and the ovalbumin group were both higher than those of the former group (P <0.05). Compared with the same group, the swelling reached the level of 7d after freezing (P <0.05). Compared with the same group after 7 days of freezing, the swelling of knee joint reduced 14 days after freezing, and the pathological score of synovial membrane decreased (P <0.05), but still higher than that at 1 day after freezing (P <0.05). Conclusion: The model of RA cold syndrome established by ovalbumin induction combined with cryogenic freezing is reliable, and the model can last at least 14 days.
