为了探讨中国仓鼠(CHA)卵丘细胞及其胞外基质(COM)和细胞外基质(ECM)对精子获能、顶体反应(AR)和受精能力的作用,以及COM中何种因子影响上述受精环节,分别采用阿利新蓝8GX法和放射免疫法(RIA)测定COM中糖胺多糖(GAG)总量和孕酮(P4)含量,并以精子穿卵试验(SPA)评价精子的受精能力。结果表明:CHA精子在COM中AR率随获能培养时间的延长而增加,呈时间依赖关系。ECM是诱发CHA精子AR的主要部位。它含P4、透明质酸(HA)和肝素(HE)等。它们均可引起CHA体外获能精子发生AR,AR率随P4和GAG剂量而增加,并存在着明显的量效关系。P4不能引起未获能精子发生AR,据此,它可以作为一种评价精子是否获能的探针。此外,P4还可明显地提高CHA精子穿卵能力。由此可得出结论,ECM中P4和GAG是引起CHA精子获能、AR和受精的因子
To investigate the effects of Chinese hamster (CHA) cumulus cells and their extracellular matrix (ECM) on sperm capacitation, acrosome reaction (AR) and fertilization ability, as well as which factors in COM affect the above The amount of glycosaminoglycan (GAG) and progesterone (P4) in COM were determined by alginate blue 8GX and radioimmunoassay (RIA), respectively. The fertilization ability of sperm was evaluated by sperm penetration test . The results showed that the AR rate of CHA sperm in COM increased with time prolonging the incubation time in a time-dependent manner. ECM is the main site of CHA induced sperm AR. It contains P4, hyaluronic acid (HA) and heparin (HE) and so on. Both of them can cause CHA in vitro spermatogenesis AR, AR rate increased with the dose of P4 and GAG, and there is a clear dose-effect relationship. P4 can not cause AR in un-enabled spermatozoa, therefore, it can be used as a kind of evaluation of sperm motility is not probable. In addition, P4 can also significantly improve CHA sperm penetration ability. From this it can be concluded that P4 and GAG in ECM are the factors that cause CHA sperm capacitation, AR and fertilization
