分别以感染复数(MOI)为0、50、100的腺病毒为载体的BMP-2基因转染体外培养的舌癌Tca8113细胞。采用荧光倒置显微镜观察转染前后Tca8113细胞形态的变化,Western blot法检测Tca8113细胞BMP-2的表达,四甲基偶氮唑盐(MTT)法检测Tca8113细胞增殖能力,并绘制生长曲线。结果:MOI为100时转染率最高,转染前后Tca8113细胞形态变化不大,Western blot检测到Tca8113细胞BMP-2的表达,转染后Tca8113细胞的增殖活性降低。结论:腺病毒介导的人BMP-2在体外能够抑制舌癌Tca8113细胞的增殖活性。
The tongue cancer Tca8113 cells cultured in vitro were transfected with adenoviral vector BMP-2 with MOI of 0,50,100 respectively. The morphological changes of Tca8113 cells before and after transfection were observed by fluorescence inverted microscope. The expression of BMP-2 in Tca8113 cells was detected by Western blot. The proliferation of Tca8113 cells was detected by MTT assay and the growth curve was drawn. Results: The transfection rate was the highest at 100 MOI. The morphology of Tca8113 cells did not change much before and after transfection. The expression of BMP-2 in Tca8113 cells was detected by Western blot. The proliferation activity of Tca8113 cells was decreased after transfection. Conclusion: Adenovirus-mediated human BMP-2 can inhibit the proliferation of tongue cancer Tca8113 cells in vitro.
