多抗甲素是一种新型的免疫增强剂,临床应用证明多抗甲素对再生障碍性贫血疗效较好。为了探讨多抗甲素治疗再障的机理,我们曾用小鼠照射后形成脾集落的方法,对造血干细胞CFU-S作了测定,发现多抗甲素有刺激造血的作用。在此基础上,我们应用电子显微镜技术,对CFU-S脾集落细胞进行了电镜观察。本文首次报告了CFU-S脾集落细胞的超微结构特征。发现多抗甲素对脾集落细胞的超微结构有明显影响,能促进脾集落细胞增殖分化,脾集落细胞显示出代谢旺盛、功能活跃、DNA及蛋白质合成增加等表现。本文根据免疫缺陷学说对再障发病机理作了探讨,从超微结构水平讨论了多抗甲素对造血干细胞的影响。
PolyA is a new type of immune enhancer, clinical application of polyA shows good effect on aplastic anemia. In order to investigate the mechanism of poly-A treatment of aplastic anemia, we used mouse irradiation to form spleen colonies method, hematopoietic stem cell CFU-S were measured and found that poly-A has the role of stimulating hematopoiesis. On this basis, we applied electron microscopy to electron microscopy of CFU-S spleen cells. This paper reports for the first time the ultrastructural features of CFU-S spleen cells. It was found that polyactin A had a significant effect on the ultrastructure of spleen cell colonies, which could promote the proliferation and differentiation of spleen colony cells. The spleen cells showed strong metabolism, active function and increased DNA and protein synthesis. In this paper, based on immunodeficiency theory of aplastic anemia on the pathogenesis was discussed from the ultrastructure of the level of discussing the effects of polyactin on hematopoietic stem cells.
