目的 分析膀胱移行细胞癌的差异表达基因 ,探讨肿瘤行为的分子生物学基础及筛选有效的肿瘤标记。方法 应用抑制性消减杂交 (SSH)方法构建在正常膀胱组织和膀胱癌组织中差异表达序列的消减文库 ,用差异筛选技术分离差异表达基因。结果 从消减文库中共获得了 9个在正常膀胱组织中高表达、在膀胱癌组织中表达下调的基因 ,分别属于细胞外基质成分和肌动蛋白细胞骨架体系。结论 细胞外基质和肌动蛋白细胞骨架体系中相关基因的表达下调可能反映了肿瘤细胞在细胞粘附、细胞间相互作用、迁移及运动等诸多过程中的异常 ,可能与癌细胞的侵袭和转移密切相关 ,对发展成为膀胱癌的分子标记物具有潜在价值 ;gelsolin、tropomyosin等抑癌基因在膀胱癌形成过程中可能起了重要作用
Objective To analyze differentially expressed genes in transitional cell carcinoma of the bladder and to explore the molecular basis of tumor behavior and to screen for effective tumor markers. Methods Suppression subtractive hybridization (SSH) was used to construct subtractive libraries of differential expression sequences in normal bladder tissues and bladder cancer tissues. Differentially expressed genes were isolated by differential screening. Results Nine genes that were overexpressed in normal bladder tissues and down-regulated in bladder cancer tissues were obtained from the subtractive library, belonging to the extracellular matrix components and the actin cytoskeleton system, respectively. Conclusion The down-regulation of the related genes in extracellular matrix and actin cytoskeleton may reflect the abnormality of tumor cells in many processes such as cell adhesion, cell-cell interaction, migration and movement, which may be related to the invasion and metastasis of cancer cells Closely related to the development of bladder cancer molecular markers have potential value; gelsolin, tropomyosin and other tumor suppressor genes in the formation of bladder cancer may play an important role
