目的探讨肠上皮下肌成纤维细胞(ISEMF)分布,表达与大肠癌转移及预后之间的关系。方法应用免疫组化方法,采用α-平滑肌肌动蛋白(а-SMA)单克隆抗体检测ISEMF在20例正常对照者大肠黏膜及80例大肠癌中的分布和表达。结果 ISEMF主要分布于正常肠粘膜中下部,呈线状分布于腺体周围,在腺体底部与粘膜肌层形成连接;在所有大肠癌肠粘膜标本中均弥漫表达,广泛分布于间质各部分。а-SMA阳性及阳性面积之间的比例在肿瘤组织及正常肠粘膜组织中的表达分别为682154.84±510027.63和263541.57±135729.21,0.1976±0.0027和0.0799±0.0302;肿瘤组织中а-SMA表达阳性明显增高,两者有显著统计学差异(P<0.01)。结论大肠癌与正常大肠粘膜间质中ISEMF的分布存在明显差异,大肠癌肠粘膜间质中ISEMF的明显增加,提示ISEMF分布与肿瘤的组织学类型及大肠癌侵袭发展有关。
Objective To investigate the relationship between the distribution and expression of intestinal epithelial fibroblasts (ISEMF) and the metastasis and prognosis of colorectal cancer. Methods The distribution and expression of ISEMF in 20 cases of colorectal mucosa and 80 cases of colorectal cancer were detected by immunohistochemistry and α-smooth muscle actin (α-SMA) monoclonal antibody. Results ISEMF mainly distributed in the middle and lower part of normal intestinal mucosa, linearly distributed around the gland, forming a connection with the muscularis mucosa at the bottom of the gland. In all of the colorectal mucosa, ISEMF was diffusely expressed and widely distributed in various parts of the stroma . The positive and positive areas of а-SMA were 682154.84 ± 510027.63 and 263541.57 ± 135729.21,0.1976 ± 0.0027 and 0.0799 ± 0.0302 respectively in tumor tissues and normal mucosa tissues. The positive expression of а-SMA in tumor tissues was significantly higher , There was a significant difference between the two (P <0.01). Conclusion The distribution of ISEMF in colorectal carcinoma and normal colorectal mucosa is obviously different. The expression of ISEMF in colorectal mucosa of colorectal carcinoma is obviously increased, which suggests that the distribution of ISEMF is related to the histological type of tumor and the invasion and development of colorectal cancer.
