为探讨同种异体尸肾移植排斥反应病人淋巴细胞CD62L、CD11a表达与T细胞亚群及CD4/CD8的关系和意义。利用单克隆抗体 流式细胞仪荧光免疫技术 ,测定 10例肾移植排斥反应病人术后不同时间外周血淋巴细胞CD62L、CD11a、CD4、CD8表达并计算CD4/CD8。结果 ,肾移植病人排斥反应时其CD62L (4 6 1± 18 7vs 31 3± 10 5 ,P <0 0 1)、CD11a (4 9 5±2 0 2vs 31 9± 12 4,P <0 0 1)、CD4(2 4 4± 7 7vs 17 9± 7 4,P <0 0 1)、CD8(14 7± 2 9vs 10 4± 3 2 ,P <0 0 5 )表达均较排斥前明显增加 ,抗排斥治疗后CD11a (14 8± 6 2vs 49 5± 2 0 2 ,P <0 0 1)、CD4(15 8± 6 4vs 2 4 4± 7 7,P <0 0 5 )和CD4/CD8(1 2 8± 0 6vs 1 73± 0 79,P <0 0 5 )均明显下降。CD62L变化和CD8呈明显正相关 (r=0 9779,P <0 0 5 )。认为淋巴细胞CD62L、CD11a、CD4、CD8表达及CD4/CD8与肾移植排斥反应密切相关。免疫抑制剂 ,尤其甲基强的松尤能明显抑制淋巴细胞CD11a、CD4表达和CD4/CD8比值可能是其发挥抗排斥作用的重要机制。
To investigate the relationship between the expressions of CD62L, CD11a, T lymphocyte subsets and CD4 / CD8 in patients with allograft rejection after cadaveric renal allograft rejection. The expression of CD62L, CD11a, CD4 and CD8 in peripheral blood lymphocytes of 10 patients with renal allograft rejection at different time after operation were measured and the ratio of CD4 / CD8 was calculated by fluorescence immunoassay using monoclonal antibody flow cytometry. As a result, CD62L (461 ± 18 7 vs 31 3 ± 10 5, P <0.01), CD11a (495 ± 202 vs 31 9 ± 12 4, P <0.01) ), CD4 (2 44 ± 7 7 vs 17 9 ± 7 4, P 0 01) and CD8 (14 7 ± 2 9 vs 10 4 ± 32, P 0 05) After anti-rejection treatment, CD11a (14 8 ± 6 2 vs 49 5 ± 202, P 0 01), CD4 (15 8 ± 6 4 vs 2447 ± 7 7, P 0 05) and CD4 / CD8 1 2 8 ± 0 6vs 1 73 ± 0 79, P 0 05) decreased significantly. The change of CD62L was positively correlated with CD8 (r = 0 9779, P <0 05). It is considered that the expressions of CD62L, CD11a, CD4, CD8 and CD4 / CD8 in lymphocytes are closely related to the renal allograft rejection. Immunosuppressive agents, especially methylprednisolone, can significantly inhibit the lymphocytes CD11a, CD4 expression and CD4 / CD8 ratio may play an important anti-rejection mechanism.
