目的:研究丙型肝炎病毒基因1b(HCV-1b)NS5A区准种变异与干扰素疗效的关系。方法:选取HCV一1b型慢性丙型肝炎患者20例,在干扰素治疗前作NS5A区准种变异等测定,并比较其与干扰素疗效的关系。结果:10例干扰素治疗有效组的单链构象多肽性(SSCP)大部分表现为2带(6例),少部分为3带(4例),平均为2.4带;而10例干扰素治疗无效组的SSCP大部分表现为3带(6例),少部分为4带(4例),平均为3.4带。两者比较有极显著差异。结论:干扰素的疗效与NS5A准种变异密切相关,有效组准种变异的程度明显低于无效组,即病人治疗前HCV准种变异较少者,HCV病毒易于被清除。采用SSCP检测HCV-NS5A准种变异以预测丙型肝炎干扰素疗效有一定的实用价值。
Objective: To study the relationship between the quasispecies mutation of NS5A gene of hepatitis C virus 1b (HCV-1b) and the efficacy of interferon. Methods: Twenty patients with HCV-1b chronic hepatitis C were selected for quasispecies variation of NS5A before interferon treatment, and their relationship with the efficacy of interferon was compared. Results: Most of the single-strand conformational polymorphism (SSCP) of 10 interferon-treated groups showed in 2 bands (6 cases) and a small proportion of 3 bands (4 cases) with an average of 2.4 bands, while 10 cases of interference Most of the SSCP patients in the treatment-ineffective group showed 3 bands (6 cases) and a small part of 4 bands (4 cases), with an average of 3.4 bands. There is a very significant difference between the two. Conclusion: The curative effect of interferon is closely related to quasispecies variation of NS5A, and the quasi-species variation of effective group is obviously lower than that of ineffective group, that is, the variation of HCV quasispecies in patients before treatment is relatively less, and the HCV virus is easy to be eliminated. The use of SSCP detection of HCV-NS5A quasispecies to predict the efficacy of hepatitis C interferon has a certain practical value.
