目的观察糖尿病动脉粥样硬化(AS)大鼠血清中二羰基化合物的表达。方法 20只GK大鼠采用左旋硝基精氨酸甲酯(L-NAME)和高脂饲料喂养法制备糖尿病AS模型(Model组),另选20只健康Wistar大鼠作为对照(NC)组。应用气相色谱-质谱联用仪(GC-MS)对大鼠血清进行分离、纯化,测定其中二羰基化合物甲基乙二醛(MG)和3-脱氧葡萄糖醛酮(3-DG)的含量。结果 Model组血清中二羰基化合物MG和3-DG含量分别为(216.7±15.6)μmol/L和(19.5±3.8)μmol/L,与NC组(85.3±24.9)μmol/L和(8.2±2.4)μmol/L相比,含量升高(P<0.05)。结论二羰基化合物含量升高可能是糖尿病AS的发病机制之一。
Objective To observe the expression of dicarbonyl compounds in the serum of diabetic rats with atherosclerosis (AS). Methods 20 GK rats were randomly divided into model group and model group, and 20 healthy Wistar rats were selected as the control group (NC group) by L-NAME and high fat diet. The rat serum was separated and purified by gas chromatography-mass spectrometry (GC-MS), and the contents of methylglyoxal (MG) and 3-deoxyglucosone (3-DG) were determined. Results The levels of dicarbonyl compounds MG and 3-DG in model group were (216.7 ± 15.6) μmol / L and (19.5 ± 3.8) μmol / L, respectively, ) μmol / L, the content increased (P <0.05). Conclusion The increase of dicarbonyl compounds may be one of the pathogenesis of diabetic AS.
