目的:探讨阴茎海绵体内注射胰岛素样生长因子1(hIGF-1)基因提高老年性大鼠阴茎勃起功能的最佳剂量和作用实效。方法:4月龄SD雄性大鼠10只(青年组);24月龄SD雄性大鼠40只,随机分为5组,每组10只:PBS对照组、hIGF-1基因10μg注射组、100μg注射组和1000μg注射组,注射后在不同时间段4周和8周行电刺激检测大鼠阴茎海绵体内压(ICP)和平均动脉压(MAP)的测定,分析比较不同剂量和不同时间hIGF-1基因治疗的效果;RT-PCR检测hIGF-1基因mRNA的表达。结果:电刺激发现老年组较青年组ICP/MAP和总ICP明显降低(P<0.05)。4和8周的治疗之后,10μg注射组、100μg注射组和1000μg注射组较PBS对照组ICP/MAP和总ICP均明显提高(P<0.05);100μg注射组和1000μg注射组在4和8周时ICP/MAP和总ICP值和青年组相仿;10μg注射组虽然不能达到和青年组相仿的治疗效果,但仍然有治疗意义;所有hIGF-1基因治疗组均可检测到hIGF-1 mRNA的表达。结论:hIGF-1基因治疗能够提高老龄大鼠的勃起功能,100μg即可达到有效的治疗,hIGF-1基因治疗至少能够维持8周时间。
Objective: To investigate the optimal dose and effect of injecting insulin-like growth factor 1 (hIGF-1) gene in penis into penis to improve penile erection in senile rats. Methods: Ten male Sprague-Dawley rats of 4 months old (youth group) and 40 male SD rats of 24 months old were randomly divided into 5 groups with 10 mice in each group: PBS control group, 10μg hIGF-1 gene injection group, 100μg Injection group and 1000μg injection group. The ICP and MAP of rat’s penis were detected by electrical stimulation at 4 and 8 weeks after injection. The levels of hIGF- 1 gene therapy; RT-PCR detection hIGF-1 gene mRNA expression. Results: Compared with the young group, the ICP / MAP and total ICP in the aged group were significantly decreased (P <0.05). After 4 and 8 weeks of treatment, the ICP / MAP and total ICP of 10μg, 100μg and 1000μg injection groups were significantly higher than that of PBS control group (P <0.05). After 100μg injection group and 1000μg injection group at 4 and 8 weeks ICP / MAP and total ICP were similar to those in youth group. Although 10μg injection group could not achieve the same therapeutic effect as young group, it still had therapeutic significance; hIGF-1 mRNA expression was detected in all hIGF-1 gene therapy groups . Conclusion: hIGF-1 gene therapy can improve the erectile function of aged rats, 100μg can achieve effective treatment, hIGF-1 gene therapy can maintain at least 8 weeks.
