按莱姆基(Lemke,1972)的制法如下: 1.取10公斤麦粒,加15公升水,在文火上煮15-20分钟。 2.煮后用笊篱捞出麦粒,摊平凉干,再拌上120克石膏和30克碳酸钙,以期调整pH,并有缓冲作用。此外,据斯托列(Stoller,1956)确认石膏尚有防止麦粒粘结,改善基质通气之效。 3.将麦粒装到容积为1公斤的奶瓶子中,装量为基容积的2/3。故而每瓶装350-400克麦粒。瓶口塞以棉塞,在温度为120℃,压力为101325帕斯卡下灭菌,历时1.5小时,灭菌后pH应为6.5-6.7, 4.俟冷至24-26℃时接种。
