用 5× 10 -6mol·L-1浓度的全反式维甲酸 (ATRA)作用于人肺鳞癌细胞株 (TUL细胞 ) ,检测细胞生长曲线 ;用MTT法检测细胞生长抑制率 ;用3H TdR掺入法测细胞DNA合成情况 ;透射电镜下观察细胞超微结构改变 ;并观察裸鼠成瘤情况。结果显示 :与对照组比较 ,经ATRA作用后 ,TUL细胞生长速度减慢 ,细胞生长曲线平坦 ,细胞生长抑制率为 42 3 % ,3H TdR掺入率明显减低 (P <0 .0 1) ,细胞DNA合成减少 ,透射电镜观察发现 ,TUL细胞呈良性分化 ,TUL细胞致瘤性明显降低。提示ATRA能抑制TUL细胞增殖 ,诱导分化 ,降低其致瘤性
The growth of human lung squamous cell carcinoma cell line (TUL) was detected by using all-trans retinoic acid (ATRA) at a concentration of 5 × 10 -6 mol·L-1. The cell growth inhibition rate was measured by MTT assay. 3H-TdR DNA synthesis was measured by incorporation method; the ultrastructure of cells was observed under transmission electron microscope; and the tumorigenesis of nude mice was observed. The results showed that compared with the control group, the growth of TUL cells was slowed down and the cell growth curve was flat after treated with ATRA. The cell growth inhibition rate was 42.3% and 3H TdR incorporation rate was significantly decreased (P <0.01) Cell DNA synthesis decreased, transmission electron microscopy showed that, TUL cells were benign, TUL cell tumorigenicity was significantly reduced. Tip ATRA can inhibit TUL cell proliferation, induce differentiation and reduce its tumorigenicity
