建立了同时测定免血清中中药马钱子所含主要生物碱——士的宁和马钱子碱的反相 HPLC 法。固定相为 Nova-Pak C_(18)(3.9×150mm,4μm)柱;流动相为乙腈-水(40:60,每1000ml 含磷酸二氢钾3.4g 及十二烷基硫酸钠1.7g);检测波长为254nm;理论塔板数按士的宁和马钱子碱计算应分别不得低于5500和3500。士的宁和马钱子碱的最低检测限分别为4.52ng 和3.24ng;二者的加样回收率分别为92.55±1.18%和90.00±1.43%。
A reversed-phase HPLC method was established for the simultaneous determination of strychnine and strychnine, the main alkaloids contained in serum-free Machizi. The stationary phase was a Nova-Pak C18 (3.9 x 150 mm, 4 μm) column; the mobile phase was acetonitrile-water (40:60, 3.4 g of potassium dihydrogen phosphate per 1000 ml and 1.7 g of sodium dodecyl sulfate); The detection wavelength is 254 nm; the number of theoretical plates calculated by strychnine and brucine should not be lower than 5500 and 3500, respectively. The minimum detection limits of strychnine and strychnine were 4.52 ng and 3.24 ng, respectively; the recovery rates of the two were 92.55±1.18% and 90.00±1.43%, respectively.
