目的:探讨龙牙楤木叶总皂苷抗肿瘤作用机理。方法:采用ELISA法检测S-180小鼠IL-2、TNF-α含量;免疫组化法检测肿瘤组织中rasP21蛋白和突变型P53蛋白含量。结果:龙牙楤木叶总皂苷明显增高S-180小鼠IL-2、TNF-α含量(P<0.05);对rasP21和P53蛋白表达明显减少(P<0.05)。结论:龙牙楤木叶总皂苷能诱生荷瘤小鼠IL-2、TNF-α含量,抑制ras癌基因蛋白P21和突变型P53蛋白的过度表达,而达到抗肿瘤的作用。
Objective: To investigate the antitumor mechanism of total saponins of Aralia elata. Methods: The levels of IL-2 and TNF-α in S-180 mice were detected by ELISA. The contents of rasP21 protein and mutant P53 protein in tumor tissues were detected by immunohistochemistry. Results: The total saponin of Aralia elata increased significantly the content of IL-2 and TNF-α in S-180 mice (P <0.05), but decreased the expressions of rasP21 and P53 (P <0.05). Conclusion: Aralia elata leaves saponin can induce tumor-bearing mice IL-2, TNF-α content, inhibit ras oncogene protein P21 and mutant P53 protein overexpression, to achieve anti-tumor effect.
