[目的]探讨阿霉素(ADM)诱导人乳腺癌细胞MCF-7上皮-间质转化的作用.[方法]体外培养人乳腺癌细胞MCF-7,用不同质量浓度ADM处理后观察形态变化,并采用细胞免疫荧光染色法检测E-cadherin和vimentin蛋白水平.[结果]ADM可使MCF-7细胞变长变扁,明显增加MCF-7细胞的vimentin蛋白水平(P<0.05),降低E-cadherin蛋白水平(P<0.05).[结论]ADM可诱导人乳腺癌细胞MCF-7上皮-间质转化.
[Objective] To explore the effect of doxorubicin (ADM) on the epithelial-mesenchymal transition in human breast cancer cell line MCF-7. [Methods] Human breast cancer cell line MCF-7 was cultured in vitro and treated with different concentrations of ADM. Morphological changes, The expression of E-cadherin and vimentin protein in MCF-7 cells was detected by immunofluorescence staining. [Results] ADM could prolong and flatten the MCF-7 cells and significantly increase the vimentin protein level in MCF-7 cells (P <0.05) cadherin protein level (P <0.05). [Conclusion] ADM can induce epithelial-mesenchymal transition in human breast cancer cell line MCF-7.
