目的:研究脓毒症时脑神经细胞凋亡的发生情况及其发病机制。方法:采用胆管内注射大肠杆菌的方法制作大鼠急性重症胆管炎模型,4天后取血化验TNFα、IL-1及IL-6活性,并取脑海马组织作原位细胞凋亡检测及电镜检查。结果:(1)观察组比对照组血IL-1、IL-6及TNFα水平明显增高。(2)用TUNEL染色法镜下观察发现,观察组的原位细胞凋亡数目较对照组增多。(3)电镜下观察发现海马区神经细胞发生早期细胞凋亡改变。结论:在大鼠发生急性重症胆管炎时,TNFα、IL-1及IL-6等细胞因子的明显增高为典型脓毒症表现,而后者可诱导脑神经细胞发生凋亡。
Objective: To study the occurrence of cerebral neuronal apoptosis and its pathogenesis in sepsis. Methods: The model of acute cholangitis in rats was established by intra-bile duct injection of E.coli. The levels of TNFα, IL-1 and IL-6 in serum were measured after 4 days. The apoptosis of hippocampus in situ was detected by electron microscopy . Results: (1) The levels of IL-1, IL-6 and TNFα in the observation group were significantly higher than those in the control group. (2) By TUNEL staining microscopy observation found that the number of apoptotic cells in situ observation group increased compared with the control group. (3) Electron microscopy showed that early apoptotic changes occurred in hippocampal neurons. Conclusion: In acute severe cholangitis, TNFα, IL-1 and IL-6 and other cytokines were significantly increased in typical sepsis patients, while the latter could induce apoptosis in brain neurons.
