目的观察藤梨根提取液对小鼠染色体DNA抗诱变作用。方法将清洁级昆明小鼠随机分为空白对照组、环磷酰胺组、低剂量I组、低剂量II组、高剂量I组、高剂量II组6组,分别予蒸馏水、低剂量藤梨根提取液、高剂量藤梨根提取液灌胃;除高剂量I组、高剂量II组外,于给药第2天予各组小鼠腹腔注射环磷酰胺(cyclophosphamide,CP)诱变,给药第4天处死各组动物。通过小鼠骨髓细胞嗜多染红细胞(polychromatic erythrocyte,PCE)微核(micronucleus,MN)的发生情况,观察藤梨根的抗诱变作用。结果低、高两种剂量的藤梨根提取液对CP诱发的小鼠MNPCE有明显的抑制作用,低剂量、高剂量组的MN发生率与环磷酰胺组比较差异有统计学意义(P<0.01,P<0.05)。低、高两种剂量的藤梨根提取液均未显示有促进小鼠体内自发MN和有抑制自发MN产生的作用。结论藤梨根提取液对诱变剂引发DNA的损伤具有抑制作用。
Objective To observe the antimutagenicity of genomic DNA extracted from ratoon root. Methods Kunming mice of clean grade were randomly divided into blank control group, cyclophosphamide group, low-dose I group, low-dose II group, high-dose I group, and high-dose II group 6 groups, respectively distilled water, low-dose vine root The extract and high-dose vine root extracts were intragastrically administered; in addition to the high-dose I group and high-dose group II, mice were intraperitoneally injected with cyclophosphamide (CP) for mutagenesis on the second day after administration. The animals were sacrificed on the fourth day of the drug. Through the occurrence of micronucleus (MN) of polychromatic erythrocyte (PCE) in mouse bone marrow cells, the anti-mutagenicity of vine root was observed. Results Low and high doses of wolfberry pear root extract significantly inhibited CP-induced MNPEC in mice. The incidence of MN in the low-dose and high-dose groups was significantly different from that in the cyclophosphamide group (P< 0.01, P < 0.05). Both low and high doses of wolfberry pear root extract did not appear to promote spontaneous MN and inhibit spontaneous MN production in mice. Conclusion The root extract of vine pear has inhibitory effect on DNA damage induced by mutagen.
