目的:观察麝香、冰片、安息香和苏合香四味芳香开窍药对脑缺血再灌注损伤大鼠的保护作用与作用机制,为该类药物开窍醒神功效与应用提供一定的药理学依据。方法:采用大鼠大脑中动脉栓塞法制备脑缺血再灌注损伤大鼠模型,4味药物在大鼠造模前连续给药3天,观察模型大鼠神经体征的变化,测定脑含水率,比色法测定血清超氧化物歧化酶(SOD)活性与丙二醛(MDA)含量,双抗体夹心酶联免疫吸附法(ELISA)测定血清肿瘤坏死因子α(TNF-α)水平。结果:与模型组比较,麝香(66.6mg/kg)、冰片(0.2g/kg)能显著降低脑含水率,提高血清SOD活性,降低血清MDA与TNF-α含量,表现出对脑缺血再灌注损伤模型大鼠的保护作用。苏合香有降低脑含水率、血清MDA与TNF-α含量,及提高SOD活性的趋势。安息香的作用则不显著。四味药物对模型大鼠的神经体征行为学评分均有一定降低趋势。结论:该类药物对脑缺血再灌注损伤大鼠有一定的保护作用,以麝香、冰片作用为优,其次苏合香,安息香作用较弱,作用机制可能与其抗氧化和抑制炎症反应有关。
OBJECTIVE: To observe the protective effect and mechanism of musk, borneol, benzoin and sylvestris aromatic resuscitation drugs on cerebral ischemia-reperfusion injury in rats, and to provide some pharmacological evidences for their efficacy and application. Methods: Rat models of cerebral ischemia-reperfusion injury were established by middle cerebral artery occlusion (MCAO). The four drugs were administered continuously for 3 days before modeling in rats. The changes of neurological signs were observed and the brain water content, Serum levels of superoxide dismutase (SOD) and malondialdehyde (MDA) were measured by colorimetric method. Serum levels of tumor necrosis factor alpha (TNF-α) were measured by double antibody sandwich enzyme-linked immunosorbent assay (ELISA) Results: Compared with model group, musk (66.6mg / kg) and borneol (0.2g / kg) significantly reduced brain water content, increased serum SOD activity, decreased serum MDA and TNF-α content, Protective effects of perfusion injury model in rats. Su-Hexiang can reduce brain water content, serum MDA and TNF-α content, and increase the trend of SOD activity. The effect of benzoin is not significant. Siwei medicine on the model rats neurological signs of behavior have a certain downward trend. CONCLUSION: These drugs have some protective effects on rats with cerebral ischemia-reperfusion injury. Musk and borneol are the best drugs, followed by the weak effect of Su-a-xiang and benzoin. The mechanism may be related to their anti-oxidation and anti-inflammation.
