目的观察血清雌二醇对系统性红斑狼疮(SLE)合并妊娠患者B淋巴细胞表面抑制性受体CD72、CD32b、白细胞相关免疫球蛋白样受体1(LAIR-1)表达的影响。方法提取40例SLE合并妊娠患者和10例健康妊娠妇女的B淋巴细胞,采集妊娠第5、6、7周外周血中雌二醇刺激B细胞进行培养。ELISA检测B淋巴细胞表面抑制性受体CD72、CD32b、LAIR-1表达水平。结果正常妊娠女性在孕5、6、7周时雌二醇浓度逐渐增加,而CD72、CD32b、LAIR-1无明显变化;分离SLE妊娠妇女B淋巴细胞,经(500、1000、1500)U/L雌二醇体外刺激后,B细胞CD72、CD32b、LAIR-1随着雌二醇剂量的增加逐渐降低,且低于正常妊娠妇女对照组。结论雌二醇可下调SLE合并妊娠患者B淋巴细胞CD72、CD32b、LAIR-1的表达。
Objective To observe the effect of serum estradiol on the expressions of CD72, CD32b and leukocyte associated immunoglobulin-like receptor 1 (LAIR-1) on B lymphatic cells in patients with systemic lupus erythematosus (SLE) complicated with pregnancy. Methods B lymphocytes were collected from 40 SLE patients with pregnancy and 10 healthy pregnant women. The estradiol-stimulated B cells were collected from the 5th, 6th and 7th weeks of pregnancy for B lymphocyte culture. ELISA was used to detect the expression of CD72, CD32b and LAIR-1 on the surface of B lymphocytes. Results The estradiol concentration of pregnant women gradually increased at the 5th, 6th and 7th week of pregnancy, while there was no significant change of CD72, CD32b and LAIR-1. The B lymphocytes of pregnant women with SLE were separated and the numbers of U (500,1000,1500) U / L estradiol stimulated the expression of CD72, CD32b and LAIR-1 in B cells gradually decreased with the increase of estradiol dosage, and was lower than that of normal pregnant women. Conclusion Estradiol can down-regulate the expression of CD72, CD32b and LAIR-1 in B lymphocytes from patients with SLE complicated with pregnancy.
