目的探讨锌和维生素E对全反式维甲酸(ATRA)诱导的类马蹄内翻足胎鼠的孕鼠胎盘中B细胞淋巴瘤-白血病2(Bcl-2)和Bcl相关X蛋白(Bax)表达的影响。方法 30只SD孕鼠随机均分为ATRA诱导组(A组,灌胃ATRA 120mg/kg)、抗凋亡干预组(B组,灌胃ATRA 120mg/kg、饲料添加维生素E 500mg/kg及饮用水添加ZnSO4·7H2O 227mg/L)和对照组(C组)。10d后剖宫产取新鲜胎盘组织,采用RT-PCR和Western blot分别检测Bcl-2、Bax mRNA和蛋白表达水平。结果与C组相比,A组Bcl-2mRNA和蛋白表达减少,Bax mRNA和蛋白表达增加(P<0.05);而B组能逆转A组各观察指标的变化过程(P<0.05)。结论锌和维生素E可能通过调节Bcl-2/Bax平衡,从而降低ATRA诱导的类马蹄内翻足胎鼠的孕鼠胎盘的凋亡作用。
Objective To investigate the effects of zinc and vitamin E on the expression of Bcl-2 and Bcl-2 in the placenta of pregnant rats induced by all-trans retinoic acid (ATRA) Impact. Methods Thirty pregnant SD rats were randomly divided into ATRA induction group (A group, ATRA 120mg / kg), anti-apoptotic intervention group (B group, intragastric administration of ATRA 120mg / kg, dietary supplementation of vitamin E 500mg / kg and drinking Water added ZnSO4 · 7H2O 227mg / L) and control group (C group). Fresh cotyledonary tissue was obtained from cesarean section 10 days later. The mRNA and protein expressions of Bcl-2 and Bax were detected by RT-PCR and Western blot respectively. Results Compared with group C, the expression of Bcl-2 mRNA and protein in group A was decreased and the expression of Bax mRNA and protein were increased in group A (P <0.05). Group B reversed the change of each index in group A (P <0.05). Conclusion Zinc and vitamin E may reduce the apoptosis of pregnant rat placenta induced by ATRA by regulating Bcl-2 / Bax balance.
