目的探讨庚型肝炎(HG)发生、发展规律及庚型肝炎病毒(HGV)的致病机理。方法用1例临床和病理确诊的单独HGVRNA阳性的慢性肝炎患者血清1次或多次分别静脉攻击中国恒河猴,观察攻击前、后动物血清学和肝组织学的动态变化,同时用HGVNS5区抗原制备的单克隆抗体进行肝脏组织免疫组化检测,并于感染后第18个月活体解剖1只雄性猴,取各种脏器观察组织学结构。结果动物感染后第30天血清ALT、AST开始升高,并持续异常达9个月之久;感染后第2个月肝组织出现点状、灶性及轻度碎屑样坏死并持续受损达18个月;HGV抗原定位呈肝细胞胞浆型;HGV感染后第18个月,除肝脏组织呈慢性病毒性肝炎改变外,其它脏器未见异常。结论HGV感染中国怛河猴初步建立,为进一步研究庚型肝炎提供了较合适的动物模型。
Objective To investigate the occurrence and development of hepatitis G (HG) and the pathogenesis of hepatitis G virus (HGV). Methods One Chinese cynomolgus monkey was intravenously challenged with 1 or more HGV RNA-positive chronic hepatitis B patients with clinically and pathologically diagnosed venous blood to observe the dynamic changes of animal serology and liver histology before and after challenge. Meanwhile, HGVNS5 Monoclonal antibody against antigen was used to detect the liver tissue immunohistochemistry. One male monkey was biopsied in the 18th month after infection, and the histology was observed with various organs. Results Serum ALT and AST began to increase at the 30th day after infection and continued to be abnormal for up to 9 months. Dot-like, focal and mild debris-like necrosis and persistent necrosis were observed in the 2nd month after infection Up to 18 months. The HGV antigen was located in the cytoplasm of hepatocytes. In the 18th month after HGV infection, no abnormalities were observed in other organs, except for the chronic viral hepatitis in the liver tissue. Conclusion HGV infection in Chinese 怛 R. monocytogenes was initially established, which provided a suitable animal model for the further study of hepatitis G.
