[目的]研究茶树EST-SSRs在山茶科植物中的通用性。[方法]选用来源于茶树EST序列的7对EST-SSRs引物,对山茶科植物的19个种进行了扩增。[结果]7对EST-SSRs引物中有5对引物能对19个供试品种进行有效扩增,通用性比率为71.43%;扩增率最高的为云南山茶凤山茶、华东山茶彩霞和云南山茶菊瓣,最低的为川滇莲蕊茶和大花杨桐;有效扩增的5对引物对中有4对引物在供试材料中表现出丰富且差异明显的多态性。[结论]从茶树基因组上开发的SSR引物在山茶科植物不同种属植物间有较高的通用性,可用于山茶科植物比较基因组研究和分析标记研究。
[Objective] The research aimed to study the commonness of EST-SSRs in Camellia plants. [Method] Seven pairs of EST-SSR primers derived from the EST sequence of tea tree were used to amplify 19 species of camellia plants. [Result] Five pairs of primers of EST-SSRs primers could effectively amplify 19 cultivars, with the ratio of 71.43%. The highest amplification rates were Yunnan camellia, Camellia sinensis and Yunnan camellia Chrysanthemum, the lowest was Chrysanthemum indicum tea and Populus dulcis Thunb. Among the five pairs of primers, four pairs of primers showed abundant and significant polymorphism in the tested materials. [Conclusion] The SSR primers developed from the genome of Camellia oleifera have higher commonality among plants of different species of theaceae and can be used for comparison genomic analysis and marker analysis of theaceae family.
