目的:采用响应面分析法优化大叶千斤拔中染料木素的提取工艺。方法:在单因素试验基础上,以提取温度、提取时间、溶媒比为响应因子,染料木素的提取率为响应值,通过对响应因子各水平的多元线性回归及二项式拟合,用响应面法选取较佳工艺,并进行预测分析。结果:各因素对染料木素提取率的影响大小依次为提取温度>提取时间>溶媒比。确定最佳提取工艺条件为提取温度80℃,提取时间3.5 h,溶媒比20 mL/g,在此条件下,大叶千斤拔中染料木素提取率为0.161 mg/g。结论:试验结果与模型预测值相符,该工艺可用于大叶千斤拔中染料木素的提取。
Objective: To optimize the extraction process of genistein by using response surface analysis. Methods: On the basis of single factor experiments, the extraction temperature, extraction time, solvent ratio as response factor and the extraction rate of genistein were used as response values. By using multiple linear regression and binomial fitting, Response surface method to select a better technology, and forecast analysis. Results: The effects of various factors on the extraction rate of genistein were as follows: extraction temperature> extraction time> solvent ratio. The optimum extraction conditions were extraction temperature 80 ℃, extraction time 3.5 h, and solvent ratio 20 mL / g. Under these conditions, the extraction rate of genistein was 0.161 mg / g. Conclusion: The experimental results are in good agreement with the predicted values of the model. The process can be used for the extraction of genistein from Ligusticum chuanxiong.
