目的:探讨益肾活血方含药血清对低糖低氧脑损伤大鼠海马神经干细胞增殖的影响。方法:从新生Wistar大鼠海马分离、培养神经干细胞(NSCs),将培养出的NSCs在95%N2,5%CO2,37℃培养6 h,低糖低氧体外模拟脑缺血损伤过程。随机分为3组,给药组给予益肾活血方15%含药血清;模型组给予正常大鼠未给药血清,体积分数为15%。正常组采用常规NSCs培养方法。各组细胞在37℃,5%CO2培养1 d后,运用MTT法、克隆计数观察损伤后NSCs的增殖情况。结果:低糖低氧脑损伤(低糖低氧损伤)后部分NSCs死亡,NSCs克隆数为(689.2±27.8)个,益肾活血方含药血清加入后克隆增大,克隆数增多,其中给药组为(738.6±26.6)个,2组比较差异有显著性(P<0.05)。MTT检测吸光度(A)模型组为0.617±0.21,给药组为0.895±0.37,2组比较差异有显著性(P<0.05)。结论:益肾活血方含药血清可促进低糖低氧脑损伤NSCs的增殖。
Objective: To investigate the effects of Yishenhuoxue Prescription on the proliferation of neural stem cells in hippocampus of hypoglycemic hypoxic-ischemic rats. Methods: Neural stem cells (NSCs) were isolated and cultured from the hippocampus of neonatal Wistar rats. The cultured NSCs were cultured in 95% N2, 5% CO2, 37 ℃ for 6 h. The hypoxic hypoxia in vitro simulated the process of cerebral ischemia. The rats in the treatment group were given 15% serum containing Yishenhuoxue decoction. The normal rats in the model group were given no serum and the volume fraction was 15%. Normal group using conventional methods of NSCs culture. After cultured at 37 ℃ and 5% CO 2 for 1 d, the proliferation of NSCs in each group was observed by MTT assay and clone counting. Results: After hypoxia-induced hypoxic-ischemic brain injury (hypoxia-hypoxia injury), some of the NSCs died, the number of NSCs cloned was (689.2 ± 27.8), the serum of Yishenhuoxue decoction added and the number of clones increased, (738.6 ± 26.6), the difference between the two groups was significant (P <0.05). MTT test absorbance (A) model group was 0.617 ± 0.21, the treatment group was 0.895 ± 0.37, the difference between the two groups was significant (P <0.05). Conclusion: Yishenhuoxue Fang containing serum can promote the proliferation of NSCs induced by hypoxia and hypoxia.
