用放射配体分析法,研究长时间失血性休克大鼠肾脏组织糖皮质激素受体(GR)变化并检测磷酯酶A2(PLA2)变化。结果显示:休克2小时后,GR的最大结合容量(R0)开始下降,并随休克时间延长而继续下降,于8小时后,GR的R0仅为对照组的21%。对糖皮质激素(GC)的亲和力也下降。同时肾脏PLA2上升,与GR的R0下降呈负相关(r=0.71.P<0.05)。说明GR功能下降所致受体水平GC功能不全对长时间失血性休克的发生、发展及与休克后肾功能损害可能有关。
Radioligand analysis was used to study the changes of glucocorticoid receptor (GR) in kidney and the changes of phospholipase A2 (PLA2) in rats after long-term hemorrhagic shock. The results showed that after 2 hours of shock, the maximum binding capacity (R0) of GR began to decrease and continued to decrease with the prolongation of shock time. After 8 hours, R0 of GR was only 21% of the control group. The affinity for glucocorticoids (GC) also decreased. At the same time, PLA2 increased in kidney, which was negatively correlated with the decrease of R0 in GR (r = 0.71, P <0.05). These results suggest that the GC level of receptor dysfunction induced by the decline of GR may be related to the occurrence and development of long-term hemorrhagic shock and the impairment of renal function after shock.
