目的通过比较S180肿瘤细胞体内、体外TLRs表达的变化,探讨TLRs在肿瘤发生、发展中的作用和意义。方法①将传代培养的S180腹水癌细胞接种于ICR小鼠腹腔左侧,建立S180腹水癌模型;②采用密度梯度离心法分离纯净肿瘤细胞;③采用半定量RT-PCR方法检测S180腹水癌体外传代培养细胞与荷瘤鼠体内瘤细胞TLR1-9 mRNA表达水平。结果TLR1、2、4、6在S180腹水癌体外传代培养细胞中表达水平高于荷瘤鼠体内瘤细胞;TLR7体外传代培养细胞中表达水平低于荷瘤鼠体内瘤细胞;TLR5、9体外传代培养细胞中有表达,荷瘤鼠体内瘤细胞无表达;TLR3、8体外传代培养细胞与荷瘤鼠体内瘤细胞均无表达。结论S180肿瘤细胞可表达TLRs,并且不同的TLRs在体内、体外的表达有所差异,提示TLRs在肿瘤的免疫逃逸中发挥一定的作用。
Objective To investigate the role and significance of TLRs in tumorigenesis and progression by comparing the expression of TLRs in S180 tumor cells in vivo and in vitro. Methods ① S180 ascites carcinoma cells were subcultured in the left side of the peritoneal cavity of ICR mice to establish S180 model of ascites carcinoma; ② the pure tumor cells were separated by density gradient centrifugation; ③ the semi-quantitative RT-PCR method was used to detect the passage of S180 ascites carcinoma in vitro Cultured cells and tumor bearing mice in vivo TLR1-9 mRNA expression levels. Results The expression level of TLR1, 2, 4, 6 in S180 ascites carcinoma in vitro was higher than that in tumor-bearing mice. The expression level of TLR7 in subcultured cells was lower than that in tumor-bearing mice. There was no expression of tumor cells in tumor-bearing mice in vivo. The cultured cells of TLR3 and 8 in vitro were not expressed in the tumor cells of tumor-bearing mice. Conclusion S180 tumor cells can express TLRs, and different TLRs expression in vivo and in vitro, suggesting that TLRs play a role in tumor immune escape.
