目的本研究旨在探讨应用siRNA下调XIAP在卵巢癌耐药细胞中的表达而逆转其对顺铂的耐药性。方法应用Western Blot、Hoechst 33258等检测卵巢癌顺铂敏感细胞株COC1和耐药株COC1/DDP中XIAP表达和卵巢癌细胞的凋亡率,并将XIAPsiRNA导入耐药细胞,比较转染前、后耐药细胞对顺铂耐药性的改变。结果卵巢癌耐药细胞COC1/DDP中XIAP蛋白表达明显比敏感细胞COC1中高(P<0.05)。XIAP siRNA可下调耐药细胞COC1/DDP中XIAP的表达,并显著增加对顺铂敏感性(P<0.05)。结论XIAP si RNA可在一定程度上逆转卵巢癌顺铂耐药。
The purpose of this study is to investigate the use of siRNA down-regulates expression of XIAP in ovarian cancer resistant cells and reverse its resistance to cisplatin. Methods Western Blot and Hoechst 33258 were used to detect the expression of XIAP and the apoptosis rate of ovarian cancer cells in cisplatin-sensitive ovarian cancer cell line COC1 and drug-resistant strain COC1 / DDP, and to transduce XIAPsiRNA into drug-resistant cells. Resistance of drug-resistant cells to cisplatin resistance changes. Results The expression of XIAP in COC1 / DDP cells was significantly higher than that in COC1 cells (P <0.05). XIAP siRNA down-regulated the expression of XIAP in COC1 / DDP cells and significantly increased the sensitivity to cisplatin (P <0.05). Conclusion XIAP si RNA can reverse cisplatin-resistant ovarian cancer to a certain extent.
