目的:研究5-氮胞苷(5-azacytidine,5-aza)对体外培养的兔骨髓间充质干细胞(mensenchymal stem cell, MSC)凋亡的影响。方法:体外分离培养兔 MSC,用不同浓度5-aza 诱导不同时间,观察 MSC 凋亡情况。结果:正常培养兔 MSC 有轻度细胞凋亡;5-aza 诱导浓度达15μmol/L 时凋亡率明显增加(P<0.01),当浓度达10μmol/L, 诱导时间延长则细胞凋亡率明显增加(P<0.05),超过15μmol/L 时出现成片细胞死亡。结论:5-aza 诱导对体外培养兔骨髓间质干细胞有诱导细胞凋亡作用,其作用程度与诱导时间及浓度有关。
AIM: To investigate the effect of 5-azacytidine (5-aza) on the apoptosis of cultured rabbit MSCs (bone marrow mesenchymal stem cells) in vitro. Methods: Rabbit MSC were isolated and cultured in vitro. The different concentrations of 5-aza were used to induce the different time. MSC apoptosis was observed. Results: The apoptotic rate of normal rabbit MSC was slightly increased. The apoptotic rate of 5-aza was significantly increased (P <0.01) when the concentration of 5-aza was 15μmol / L, Increased (P <0.05), and over 15 μmol / L resulted in sheet-forming cell death. CONCLUSION: 5-aza can induce the apoptosis of cultured rabbit bone marrow mesenchymal stem cells in vitro. The degree of action is related to the induction time and concentration.
