从建立大鼠前列腺上皮细胞cDNA文库入手,获得大鼠细胞角蛋白8(CK8)cDNA克隆及其全部核苷酸排列顺序,探讨雄激素对该基因mRNA表达的影响。发现去睾大鼠前列腺和精囊CK8mRNA表达增强5~10倍,且持续23d以上。雄激素可抑制其过度表达,雌激素无效;雄激素拮抗剂可消除雄激素的抑制作用,提示前列腺CK8基因是一类新的雄激素抑制性基因,该基因可能经由雄激素受体机制而起调节作用。
Starting from the establishment of rat prostate epithelial cell cDNA library, we obtained the rat cytokeratin 8 (CK8) cDNA clone and its nucleotide sequence, and investigated the effect of androgen on mRNA expression of this gene. The results showed that the expression of CK8mRNA in prostate and seminal vesicle of testis was increased 5 ~ 10 times and lasted for more than 23 days. Androgen can inhibit its overexpression, estrogen is invalid; Androgen antagonist can eliminate the inhibitory effect of androgen, suggesting that prostate CK8 gene is a new type of androgen inhibitory gene, the gene may be via the androgen receptor mechanism Regulatory effect.
