目的探讨糖尿病鼠肾脏是否存在低氧状态及其与糖尿病肾病(DN)的关系。方法将雄性SD大鼠随机分为DN组和正常对照(NC)组,采用链脲佐菌素诱导DN大鼠模型,连续多时点检测尿白蛋白、血糖、血尿素和血肌酐,同时行肾脏常规病理检查,免疫组化检测肾脏缺氧诱导因子1α(HIF-1α)与血红素氧化酶1(HO-1)的表达变化,并分析其相关性。结果 HIF-1α和HO-1在NC组肾小管仅轻度表达;而在DN组表达明显增强(P<0.01),主要集中在皮质区和皮髓质交界区的肾小管,峰值在8~12周。HIF-1α和HO-1表达变化与尿白蛋白、肾脏肥大指数和肾小球体积呈正相关。结论糖尿病肾脏缺氧状态参与了DN的发生发展。
Objective To investigate the hypoxia status of diabetic rat kidney and its relationship with diabetic nephropathy (DN). Methods Male Sprague-Dawley rats were randomly divided into DN group and normal control (NC) group. DN rats were induced by streptozotocin (STZ). Urinary albumin, blood glucose, blood urea nitrogen and serum creatinine were detected at different time points. The expression of hypoxia inducible factor 1α (HIF-1α) and heme oxygenase 1 (HO-1) in kidney were detected by routine pathological examination and immunohistochemistry. The correlation was also analyzed. Results The expression of HIF-1α and HO-1 was only mildly expressed in the renal tubules of the NC group, but significantly increased in the DN group (P <0.01). The HIF-1α and HO-1 mainly localized in the renal tubules in the cortex and cortex, 12 weeks. The expressions of HIF-1α and HO-1 were positively correlated with urinary albumin, renal hypertrophy index and glomerular volume. Conclusion Diabetic kidney hypoxia is involved in the occurrence and development of DN.
