目的利用RNA干扰(RNAi)技术抑制survivin基因的体内表达,探讨survivin沉默对结直肠癌裸鼠移植瘤生长的抑制作用。方法将survivin-siRNA重组腺病毒载体pBAsi-survivin导入裸鼠皮下的移植瘤内,观察不同时间点对肿瘤生长的抑制作用以及肿瘤的抑制率。同时用RT-PCR方法检测干预组(成功转染质粒组),对照组(空载体对照组)和空白对照组(生理盐水)中survivin mRNA表达情况。Western印迹检测survivin蛋白的表达。结果与对照组相比,survivin-siRNA重组载体导入裸鼠皮下移植瘤后,肿瘤体积增长缓慢,随着时间的延长,肿瘤生长抑制作用明显,survivin mRNA表达明显降低,survivin蛋白表达下降。结论 survivin-siRNA重组载体能显著抑制结直肠癌裸鼠移植瘤的增殖,Survivin mRNA和蛋白表达明显降低,不同时间点肿瘤抑制率不同,且随时间呈正相关。
Objective To inhibit the expression of survivin gene by RNA interference (RNAi) technique in vivo and to investigate the inhibitory effect of survivin silencing on the growth of xenografted colorectal cancer in nude mice. Methods The survivin-siRNA recombinant adenovirus vector pBAsi-survivin was transplanted subcutaneously in nude mice. The inhibitory effect on tumor growth and tumor inhibition at different time points were observed. At the same time, the expression of survivin mRNA in intervention group (plasmid transfected group), control group (empty vector control group) and blank control group (saline) were detected by RT-PCR. Western blotting was used to detect the expression of survivin protein. Results Compared with the control group, the tumor volume was slowly increased after the survivin-siRNA recombinant vector was transplanted into the nude mice subcutaneously. The tumor growth inhibition was obvious, the expression of survivin mRNA was significantly decreased and the expression of survivin protein was decreased. Conclusion The survivin-siRNA recombinant plasmid can significantly inhibit the proliferation of colorectal cancer xenografts in nude mice, and the expression of Survivin mRNA and protein is significantly decreased. The tumor inhibition rates at different time points are different and positively correlated with time.
