目的观察艾可合剂(AKHJ)抗实验性急性前列腺炎的作用及急性毒性反应,考察其有效性和安全性。方法用1%角叉菜胶生理盐水溶液致大鼠急性前列腺炎模型,观察AKHJ抗实验性急性前列腺炎作用。设AKHJ高、中、低3个剂量组,每日分别给予AKHJ(含生药26.0,13.0,6.5 g/kg)。连续灌胃7 d。模型对照组和假手术组按同法每日灌胃等量蒸馏水。另设最大给药量组和生理盐水对照组,采用灌胃给药。结果服用AKHJ后,模型大鼠前列腺质量不同程度减轻;光学显微镜显示腺腔内炎症细胞浸润程度及纤维母细胞增生程度有所减轻,高、中剂量组较模型对照组差别有统计学意义(P<0.01)。按生药量计算小鼠对AKHJ最大耐受量>156.4 g/kg(生药/体质量),相当于临床给药剂量的20倍。实验期间小鼠活动略减少,未出现中毒症状,AKHJ最大给药量组和生理盐水组小鼠均无死亡,体质量增加相近。结论AKHJ具有良好的抗急性前列腺炎作用,且毒性小,安全性高。
Objective To observe the effect and acute toxicity of AKHJ on experimental acute prostatitis, and to investigate its efficacy and safety. Methods Acute prostatitis model was induced by 1% carrageenin saline solution. The effect of AKHJ on experimental acute prostatitis was observed. The AKHJ high, middle and low dose groups were given daily AKHJ (containing crude drugs 26.0, 13.0, 6.5 g/kg). Continuous gavage 7 d. The model control group and the sham operation group were given the same amount of distilled water per day by the same method. Separately, the maximum dose group and the saline control group were administered by gavage. Results After taking AKHJ, the prostate quality of the model rats was relieved to varying degrees; light microscopy showed that the degree of inflammatory cell infiltration and fibroblast proliferation in the glandular cavity was reduced, and the difference between the high and middle dose groups was statistically significant compared with the model control group (P <0.01). The maximal tolerable dose of AKHJ in mice was calculated to be >156.4 g/kg (crude drug/body weight), equivalent to 20 times the clinical dose. During the experiment, the mouse activity decreased slightly, and no symptoms of poisoning occurred. No death occurred in the AKHJ maximal dose group and in the saline group, and the body mass increased similarly. Conclusion AKHJ has good anti-acute prostatitis effect, and has low toxicity and high safety.
