目的:研究蜀葵花总黄酮提取工艺及含量测定的最佳方法。方法:利用比色法测定蜀葵花中总黄酮含量,以总黄酮提取率为考察指标,利用正交试验设计考察乙醇体积分数、料液比、提取时间及提取温度对蜀葵花总黄酮提取工艺的影响。同时,比较不同测定方法所测得的结果。结果:蜀葵花总黄酮最佳提取工艺为:乙醇体积分数为60%、料液比为1:12、提取时间为2h、提取温度为90℃。不同测定方法测定的结果差异不大。结论:醋酸-醋酸钠缓冲溶液和AlCl3显色的方法及提取工艺简单、稳定、可行,为蜀葵花总黄酮的开发应用提供了依据。
Objective: To study the extraction of total flavonoids of Hollyhock flower and the best method to determine its content. Methods: The content of total flavonoids in hollyhock flower was determined by colorimetric method. The extraction rate of total flavonoids was taken as the index. The orthogonal experiment was used to study the effects of ethanol volume fraction, solid-liquid ratio, extraction time and extraction temperature on the extraction of total flavonoids influences. At the same time, compare the results of different measurement methods. Results: The optimal extraction technology of total flavonoids of hollyhock was as follows: the volume fraction of ethanol was 60%, the solid-liquid ratio was 1:12, the extraction time was 2h and the extraction temperature was 90 ℃. The results of different measurement methods were not significantly different. CONCLUSION: The method of acetic acid-sodium acetate buffer solution and AlCl3 coloration and the extraction process are simple, stable and feasible, which provide the basis for the development and application of the total flavonoids of hollyhock flower.
