使用热灭活的卡介苗刺激培养的人肺腺上皮细胞(SPC-A-1),利用逆转录聚合酶链反应(RT-PCR)法和Northern杂交分析,证实了卡介苗能诱导SPC-A-1细胞β-防御素-1基因(hBD-1)的增强表达。卡介苗菌量和刺激时间与SPC-A-1细胞hBD-1基因表达量的关系曲线显示,hBD-1 mRNA 在人肺腺上皮细胞的增强表达在一定范围内表现为具刺激剂量和时间依赖关系。实验为开发增强粘膜抗菌肽基因表达的免疫增强剂以防治粘膜感染的研究打下了基础。
Cultured human lung adenocarcinoma cells (SPC-A-1) were stimulated with heat-inactivated BCG, and the results showed that BCG could induce the apoptosis of SPC-A-1 cells by using reverse transcriptase polymerase chain reaction Enhanced expression of the cell beta-defensin-1 gene (hBD-1). The curve of the relationship between the amount of BCG and the stimulation time and the expression of hBD-1 in SPC-A-1 cells showed that the enhanced expression of hBD-1 mRNA in human lung glandular epithelial cells showed dose- and time-dependent effects in a certain range . The experiment lays the foundation for the development of immunostimulants that enhance the expression of mucosal antimicrobial peptide genes to prevent and treat mucosal infections.
