目的探讨连接蛋白43(Cx43)在培养的新生大鼠心室肌细胞中的时空表达变化规律。方法利用免疫细胞化学方法及免疫电镜技术,观测培养2、4、8、10、12、16、20、26、30d大鼠心室肌细胞Cx43蛋白的表达;结果培养第2d心肌细胞开始表达Cx43,表达部位主要在细胞质中,呈点状;第4d细胞质中点状分布减少,主要集中在细胞膜连接处;第10d在细胞连接处增多,呈条、链状分布;以后基本恒定,无明显变化;第30d时Cx43颗粒出现紊乱分布;结论Cx43在培养的新生大鼠心肌细胞中的表达随培养时间延长而出现位置和量的变化,与培养心肌细胞的生长、发育及功能变化一致。
Objective To investigate the spatial and temporal changes of connexin 43 (Cx43) in cultured neonatal rat ventricular myocytes. Methods Immunocytochemistry and immunoelectron microscopy were used to observe the expression of Cx43 protein in ventricular myocytes of rats at 2, 4, 8, 10, 12, 16, 20, 26 and 30 days after culture. Results Cardiomyocytes began to express Cx43 on day 2, The expression sites were mainly in the cytoplasm, showing dot-like shape. The distribution of punctate in the cytoplasm decreased on the 4th day, mainly in the cell membrane junctions. On the 10th day, The distribution of Cx43 particles appeared disorganized on the 30th day. Conclusion The expression of Cx43 in cultured neonatal rat cardiomyocytes changed with the time of culture, which was consistent with the growth, development and function of cultured cardiomyocytes.
