以红阳猕猴桃叶片、嫩茎、花蕾为外植体,MS为基本培养基,设置不同外植体消毒方法、不同的激素配方进行组织培养,比较了不同消毒剂对外植体的消毒效果、愈伤组织、不定芽的诱导情况,以及形成愈伤组织能力较强的外植体种类。结果表明:用75%酒精处理外植体30s,再以1%NaClO消毒10min,消毒效果达75.0%;诱导愈伤组织最适培养基为MS+1.0mg/L ZT+0.1mg/L NAA;诱导不定芽最适培养基为MS+1.0mg/L 6-BA+0.1mg/L NAA;用嫩茎作为外植体,诱导愈伤组织效果最好。
In this experiment, the leaves, tender stems and buds of Actinidia sagittatum were used as explants and MS as the basic medium. Different explants were disinfected and different hormone formulas were used for tissue culture. The disinfection effect of different disinfectants on the explants was compared. Injury, adventitious bud induction, and the formation of callus more capable explant species. The results showed that: the explants were treated with 75% alcohol for 30s and then disinfected with 1% NaClO for 10min, the disinfection effect reached 75.0%. The optimum medium for inducing callus was MS + 1.0mg / L ZT + 0.1mg / L NAA. The best medium for inducing adventitious buds was MS + 1.0mg / L 6-BA + 0.1mg / L NAA.
