Radioactive enzyme assay (Radioenzymatic assay) is one of the most sensitive and accurate method for the determination of catecholamines. It is used to study the changes of catecholamines in plasma or tissue under physiological and pathological conditions. The principle of determination was that S-adenosine [~ 3H] methylmethionine ([~ 3H] SAM) was used as a methyl donor and catalyzed by catechol-O-methyltransferase (COMT) To convert [~ 3H] methyl derivatives of dopamine (DA), norepinephrine (NA) and epinephrine (Ad) into 3- [~ 3H] Methyl norepinephrine ([~ 3H] NMN), 3-O- [~ 3H] adrenaline ([~ 3H] MN). After extraction, TLC separation and liquid scintillation counting, the contents of DA, NA and Ad in plasma or tissue were determined. near
