目的:建立一种荧光标记ABO基因分型的方法。方法:利用新设计的引物,在引物5′端标记荧光,经PCR扩增后用ABI3100遗传分析仪电泳。结果:所有样本扩增出相应产物,基因分型结果与血清学一致。结论:本研究采用荧光标记引物PCR-SSP法进行ABO基因分型,灵敏度高、操作简单、结果准确,为ABO基因分型提供了新的方法。
Objective: To establish a fluorescent ABO genotyping method. Methods: The newly designed primers were used to label the 5 ’end of the primers for fluorescence. After amplification by PCR, they were electrophoresed by ABI3100 genetic analyzer. Results: The corresponding products were amplified from all the samples and the results of genotyping were consistent with those of serology. Conclusion: In our study, PCR-SSP method was used to detect ABO genotype. The sensitivity, simple operation and accurate results provided a new method for ABO genotyping.
