Ⅲ型干扰素(interferon,IFN)由包含IFN-λ1,IFN-λ2,IFN-λ3三种IFN-λ亚型在内的细胞因子组成。与I型和Ⅱ型干扰素类似,Ⅲ型干扰素不仅具有抗病毒及抗肿瘤免疫效应,还在固有免疫及适应性免疫中发挥了重要作用。我们研究了miR-150在Poly(I:C)活化Toll样受体3(toll-like receptor-3,TLR3)通路后分泌IFN-λ过程中的作用。实验选用10~12周的C57/BL6小鼠和miR-150基因缺陷小鼠,尾静脉注射Poly(I:C),4h后处死小鼠并取血收集血清,检测血清中IFN-λ水平的变化。结果显示在注射Poly(I:C)后,miR-150基因缺陷小鼠血清IFN-λ水平显著低于C57/BL6小鼠,提示miR-150缺陷对Poly(I:C)活化TLR3通路后分泌IFN-λ起抑制作用。miR-150可能促进TLR3通路IFN-λ的分泌。
Type III interferon (IFN) is composed of cytokines including IFN-λ1, IFN-λ2 and IFN-λ3 subtypes. Like type I and type II interferons, type III interferons not only have antiviral and antitumor immune effects but also play an important role in innate immunity and adaptive immunity. We investigated the role of miR-150 in the secretion of IFN-λ following Poly (I: C) activation of the toll-like receptor-3 (TLR3) pathway. The experiment selected C57 / BL6 mice and miR-150 gene-deficient mice for 10-12 weeks. Poly (I) C was injected into the tail vein. After 4 hours, the mice were sacrificed and serum was collected to detect the level of IFN- Variety. The results showed that the level of IFN-λ in miR-150-deficient mice was significantly lower than that in C57 / BL6 mice after Poly (I: C) injection, suggesting that miR-150 deficiency secreted Poly (I: C) IFN-λ plays a suppressive role. miR-150 may promote the secretion of TLR3 pathway IFN-λ.
