青霉素类药物的薄层层析的检出试剂,具有特异性差,或虽有特异性但不够灵敏,且操作不简便的缺点。本文报导了应用氯铂酸与青霉素中的硫醚基反应,可作为一灵敏而有特异性的试剂,快速简便地检出薄层层析中各种青霉素及与青霉素有关的物质。测定时样品液需新鲜配备,将样品溶解于水、缓冲液或有机溶媒,配制成10或100微克毫升的溶液,准确点样1~10微升于硅胶G或纤维素薄板上,按下述溶剂系统进行单向薄层层析:(1)正丁醇-乙醇-水(12∶8∶2);(2)正丁醇-乙醇-乙酸-水(50∶15∶15∶20);(3)正丁醇-乙酸-水(60∶15∶25),展开10厘米距离,待层析板完全干燥后,以氯铂酸试剂喷雾检出斑点。
