目的 研究胶原蛋白酶解物 (CEH)对大鼠血压、豚鼠肠平滑肌及猪肺ACE的作用。方法 选用正常麻醉大鼠、SHR及肾性高血压大鼠 ,采用颈动脉插管法检测CEH对血压的影响 ;CEH对离体豚鼠回肠平滑肌的作用。结果 CEH对猪肺血管紧张素转换酶 (ACE)的IC50 为 2 6μmol/L。 1.5g/kg静脉注射能降低正常麻醉大鼠和SHR大鼠的血压 ,1.0g/kg静脉注射对肾性高血压大鼠也引起降压效应。单次口服 2g/kgCEH ,正常及SHR大鼠血压无明显变化 ,连续应用 ( 2g/kg) 15d ,SHR大鼠则呈现弱的降压效应。 7.5g/lCEH对离体豚鼠回肠平滑肌产生兴奋收缩效应。结论 CEH的降压作用和兴奋豚鼠平滑肌的作用可能与抑制ACE有关
Objective To study the effect of collagen hydrolyzate (CEH) on blood pressure, guinea pig intestinal smooth muscle and porcine lung ACE. Methods The normal anesthetized rats, SHR and renal hypertensive rats were selected. The effect of CEH on blood pressure was measured by carotid artery cannulation. The effect of CEH on isolated rat ileum smooth muscle was studied. Results The IC50 of CEH to porcine pulmonary angiotensin converting enzyme (ACE) was 26 μmol / L. 1.5g / kg intravenous injection can reduce the normal anesthetized rats and SHR rats blood pressure, 1.0g / kg intravenous injection of renal hypertension also cause antihypertensive effect. In a single oral administration of 2 g / kg CEH, there was no significant change in blood pressure of normal and SHR rats. Continuous administration (2 g / kg) for 15 days resulted in a weak antihypertensive effect in SHR rats. 7.5g / l CEH excitatory contraction of isolated guinea pig ileum smooth muscle. Conclusions The antihypertensive effect of CEH and the excitement of smooth muscle in guinea pigs may be related to the inhibition of ACE
