目的:了解芪竹方对细胞的毒作用,了解芪竹方对人脐静脉内皮细胞(HUVEC)刺激增殖后的细胞内蛋白酪氨酸激酶(PTK)活性和P-STAT3表达的影响。方法:建立对比模型组,用CCK-8法检测芪竹方对HUVEC增殖的影响,用ELISA法定量测定细胞中PTK含量,用Western Blot法测定细胞P-STAT3表达。结果:芪竹方对HUVEC正常增殖无影响;HUVEC被酸性成纤维细胞生长因子(aFGF)刺激后,空白组PTK和P-STAT3表达异常,芪竹方组对PTK表达有抑制作用,抑制率达65%,同时对P-STAT3表达有抑制,高浓度的芪竹方组作用与阳性药相当。结论:芪竹方对细胞无毒作用,对细胞的PTK和P-STAT3的异常表达有抑制作用。
Objective: To investigate the cytotoxic effect of Qizhu Recipe on cells and investigate the effect of Qizhu Recipe on the activity of protein tyrosine kinase (PTK) and the expression of P-STAT3 in human umbilical vein endothelial cells (HUVECs). Methods: The model group was established. The effect of Qizhu Decoction on the proliferation of HUVEC was detected by CCK-8 method. The PTK content was determined by ELISA and the expression of P-STAT3 by Western Blot. Results: Qi-Zhu-Fang had no effect on the normal proliferation of HUVEC. The expression of PTK and P-STAT3 in the blank group was abnormal after the HUVEC was stimulated by aFGF, while the inhibition rate was 65%, at the same time the inhibition of P-STAT3 expression, the high concentration of Qizhu Fang group and positive drugs. Conclusion: Qizhu Recipe has no toxic effect on cells and inhibits the abnormal expression of PTK and P-STAT3.
