【目的】探讨比较中药连黛片及其主要单体成分在诱导癌细胞凋亡及引起DNA损伤方面的不同药理作用。【方法】用复方连黛片的大鼠药物血清 ,以及黄连的主要药效成分小檗碱、青黛的主要药效成分靛玉红分别作用于人胃癌细胞株MGC 80 3 ,观察药物对细胞生长和凋亡的影响 ,及其对细胞DNA损伤的作用。【结果】连黛片血清高低剂量组及小檗碱各剂量组细胞与相应对照组细胞生长形态相比较差异较大 ;甲基绿—派诺宁染色后 ,细胞表现出典型的凋亡形态 ;对细胞生长率均有明显抑制。靛玉红各浓度组对细胞生长抑制不明显。琼脂糖凝胶电泳分析表明 :小檗碱各浓度组可使细胞DNA裂成大片段 ;连黛片血清各浓度组对MGC 80 3细胞染色质DNA的损伤作用较小檗碱各浓度组小 ;靛玉红在实验浓度下对细胞的DNA无损伤作用。【结论】中药连黛片大鼠药物血清与小檗碱一样均能抑制癌细胞生长及诱导细胞凋亡 ,但其作用比小檗碱弱 ,对DNA的损伤程度也较轻 ,这可能是连黛片血清组中由于血清影响了细胞对小檗碱的吸收所致
【Objective】 To explore the different pharmacological effects of the traditional Chinese medicine Lianqi tablet and its monomeric components in inducing apoptosis of cancer cells and causing DNA damage. 【Methods】 The rat drug serum of Fufang Lianqi Tablets, and the main medicinal components of Berberine, indole red, acted on the human gastric cancer cell line MGC 80 3 and observed the growth of the drug. And the effect of apoptosis, and its effect on cellular DNA damage. 【Results】 Compared with the control group, the cell growth morphology of Lianbai Tablets serum high-low dose group and berberine groups of each dose group had a greater difference; after methyl green-Pyronin staining, the cells showed a typical apoptotic morphology; The cell growth rate was significantly inhibited. Indirubin concentrations did not significantly inhibit cell growth in each concentration group. Agarose gel electrophoresis analysis showed that the berberine concentration in each group could split the DNA into large fragments, and the concentration of sulfoxide in each concentration group had less effect on the damage of chromatin DNA in MGC 80 3 cells. Indirubin has no damage to DNA of cells under experimental concentration. 【Conclusion】 The serum of traditional Chinese medicine Lianqi Tablets can inhibit the growth of cancer cells and induce cell apoptosis similarly to berberine, but its effect is weaker than that of berberine, and the degree of DNA damage is also lighter.
