介绍目前常用的双歧杆菌感受态细胞制备方法,并系统研究影响双歧杆菌电转化效率的关键因素。通过研究,菌体在A600为0.3~0.5时收集以制备感受态细胞,制备好的感受态细胞应尽早用于电转化;最佳电场强度为12.5 kV/cm;转化后的细胞复苏培养2 h为佳。感受态细胞的转化效率可达103CFU/μg DNA。
This paper introduces the current methods of preparation of competent cells of Bifidobacterium and systematically studies the key factors affecting the efficiency of Bifidobacterium electrotransformation. Through the study, the cells were collected when the A600 was 0.3-0.5 to prepare competent cells, and the prepared competent cells should be used for electrotransformation as early as possible. The best electric field intensity was 12.5 kV / cm. The transformed cells were resuscitated for 2 h Better. The transformation efficiency of competent cells can reach 103CFU / μg DNA.
