目的探讨猕猴模拟失重后脊髓中c-fos基因mRNA和蛋白表达的变化。方法选用15只雄性猕猴,随机分为3组:模拟失重组,模拟失重后恢复组以及对照组。通过HE染色法观察脊髓颈膨大及腰膨大组织学的改变,通过免疫组织化学染色及实时荧光定量PCR(Q-PCR)技术检测3组猕猴脊髓颈膨大及腰膨大中c-fos基因的表达情况。结果模拟失重组及失重后恢复组脊髓颈膨大及腰膨大神经元较对照组皱缩,组织间隙增宽,恢复组较失重组有所减轻;FOS蛋白在脊髓颈膨大及腰膨大3组中均呈阳性表达,主要表达于神经胶质细胞;失重组及恢复组中脊髓颈膨大和腰膨大c-fos mRNA表达量较对照组有所增加,但差异无明显统计学意义。结论模拟失重引起猕猴颈段及腰段脊髓一定程度的组织学损伤,终止负荷后有所恢复,对c-fos基因的表达无明显影响。
Objective To investigate the changes of c-fos mRNA and protein expression in the spinal cord of cynomolgus monkeys after simulated weightlessness. Methods Fifteen male macaque monkeys were randomly divided into three groups: simulated weightlessness group, simulated weightlessness recovery group and control group. The changes of cervical enlargement and lumbosacral histology were observed by HE staining. The expression of c-fos gene was detected by immunohistochemical staining and real-time fluorescent quantitative PCR (Q-PCR) . Results Compared with the control group, the enlargement of the medullary cervical enlargement and lumbar enlargement of the neurons in the simulated weightlessness group and the weightlessness recovery group were broadened, and the recovery of the group was less than that in the weightless group. The expression of FOS protein in the cervical spinal neck enlargement and lumbar enlargement The expression of c-fos mRNA in spinal cord neck enlargement and lumbar enlargement in the weightlessness and recovery groups was higher than that in the control group, but the difference was not statistically significant. Conclusion Simulated weightlessness can cause certain degree of histological damage in the cervical and lumbar spinal cord of the rhesus macaques. After the termination of the load, it recovered and had no significant effect on the expression of c-fos gene.
